Blotting hybridization of cloned petunia DNA fragments showed that a 3250bp restriction fragment hybridizes with the pea chalcone synthase (CHS) gene. This 3250bp band was isolated from a gel and further digested with either PstI or HindIII, as shown in a) below. To more precisely locate the petunia CHS gene in this 3250bp region, the gel was blotted onto a nylon membrane and probed with the pea CHS probe. The band patterns as visualized in autoradiography are shown in b). From this information, construct a restriction map of the 3250bp fragment, showing sites for PstI, HindIII, and the approximate location of the petunia CHS gene.
