Instrument
Circular dichroism is most commonly used in biochemistry to measure protein secondary structure but it can also probe protein tertiary structure and nucleic acid structure. Our NSERC-funded Jasco J810 Circular Dishroism Spectropolarimeter-Fluorometer can measure circular dichroism, absorption, and fluorescence from 170 nm to 1100 nm. It is equipped with a Julabo circulating water bath and a Peltier thermostatted cell holder with stirrer for temperature control. The instrument allows concurrent measurements of all 3 phenomena so, for example, it is possible to measure far UV CD and fluorescence during thermal melting of a protein.
